Instrument: Gene Navigator® System

Pulsed-field gel electrophoresis (Pulsed-field gel electrophoresis, PFGE) has been developed for the electrophoretic separation of large molecules in agarose gel. In this method, pulsed, wavy and vertical electric fields are applied to the gel. The larger the DNA molecule, the longer it takes to reorientate. In this way, they are separated according to their size.

Applications

PFGE separates DNA fragments from 20 kb to 10 Mb in length.

The largest chromosome ever separated by the Gene Navigator system comes from a fungus and is 12.6 million base pairs.

Specifications

  • The system has a hexagonal electrode kit that highlights the running sequence.
  • HEX electrodes producing homogeneous waves can be used for optimal separation.
  • It is easy to program with nine running procedures, each containing six running phases.
  • It allows evenly distributed temperature and constant pH control for repeatable and specific running conditions.
  • The self-sealing gel loader can be used to load gels up to 1 cm thick.

Sample Requirements

Liquid or solid samples can be loaded into the wells. Usually, 0.5-1.0 µg of DNA is loaded into each well.
